Test of the esculinase
The esculine is a formed heteroside of a complex glucid which can be degraded by the esculinase. The products of the reaction are the Glucose and the Esculétine. The formed esculétine can react with the ions Fer III to give a black precipitate.

If the medium is black: iron III reacted with the esculétine; the bacterium has the enzyme esculinase
- If the medium remains gray, it did not have reaction there; the bacterium does not have the enzyme
