Cyclo-oxygénase

The cyclo-oxygénase (COX) is a Enzyme which allows the formation of Prostaglandine S starting from the arachidonic Acide. Its action is inhibited by the anti-inflammatory drugs not stéroïdiens (AINS) like the aspirine.
Physiology
See also Prostaglandin and Eïcosanoïde for more detailsCOX belongs to a complex of enzymes which converts the arachidonic Acide into prostaglandin H2 (PGH2), the precursor of all the prostanoïdes. The complex consists of a isoenzyme COX and a Peroxydase. Currently one knows three Isoenzyme S COX : COX-1 , COX-2 and COX-3 . It is a system cyclopentane with two arms.
COX-1 and COX-2 . Although both enzymes act basically in the same fashion, selective inhibition edge make has difference in terms off side-effects. COX-1 is considered has constitutive enzyme, being found in most mammalian concealments. COX-2 , one the other hand, is undetectable in most normal tissues. Pharmacology
" COX-1" belong to a family of enzymes constitutive of the organization, which are in a vast majority of the cells, and implied in many mechanisms. Their activity is indirectly related to the production of the thromboxanes. " COX-2" belong to a family of inductive enzymes by multiple pro-inflammatory factors (cytokines, let us interfere…). The enzymatic activity of " type; COX-2" is expressed in the vascular, renal cells endothéliales and even on the level of the central nervous system. It is noted that they contribute to the good performance of the organization if they are well controlled. They are as a persons in charge majority of the inflammatory activity, by a surexpression of the " COX-2".
Traditional AINS
Aspirine, ibuprofene, nifluril. They aim primarily the " COX-2" by inhibiting them. If they have a bad selectivity, they can cause side effects which are related to the inhibition of the " COX-1".
