Columbia with blood
The Columbia with blood is a Culture medium and insulation of demanding bacteria not needing particular factors like Streptococus .
Composition
- special Mixture of Peptone S: 23 grams,
- Amidon: 1 G,
- Sodium chloride: 5 G,
- agar: 10 G,
- Blood: 50 ml.
The Potentiel hydrogen (pH) is of 7,3.
Preparation
42,5 G per liter. Traditional sterilization. Blood is added fruitlessly in the sterile medium in superfusion. One can carry out a gélose with blood cooks if the medium is maintained with 75-80 °C.
Reading
The Hémolyse being destruction of the red globules:- colonies surrounded by a clear halation: béta-hémolyse (hémolyse total);
- colonies surrounded by a greenish halation: alpha-hémolyse (hémolyse partial);
- other colonies: gamma-hémolyse (absence of hémolyse).
